Calculating HED

The calculation of HED is based on the principle of allometry, which describes the relationship between body size and metabolic rate across species. Allometric scaling is used to account for the differences in body weight between animals and humans. 

This involves taking the dose given to an animal and adjusting it to account for the differences in body weight between the animal and an average adult human. The HED is calculated using the following formula (Human Equivalent Dose Calculator):

HED (mg/kg) = animal dose (mg/kg) x (animal weight (kg) / human weight (kg))^0.33

For example, if a drug is administered to a rat at a dose of 10 mg/kg and the average weight of a rat is 0.25 kg, and the average weight of an adult human is 60 kg, the HED would be:

HED (mg/kg) = 10 mg/kg x (0.25 kg / 60 kg)^0.33

HED = 0.53 mg/kg

This calculation estimates the equivalent dose that would be required to achieve similar effects in humans as observed in rats. It is important to note that the HED is only an estimation and may not always accurately predict the appropriate dose for humans.

Importance of HED in Drug Development

HED is an important tool in drug development, particularly in the early stages of clinical trials. Animal studies are often used to determine the safety and efficacy of a drug before it is tested in humans. However, animal studies cannot account for all of the differences in physiology and metabolism between animals and humans. The use of HED can help researchers estimate a safe and effective dosage range for human subjects based on the results of animal studies.
